Fragment-based lead design

Abstract: State-of-the-art approaches to the fragment-based design of organic compounds with desired properties are considered. The review covers methods, which are used in different steps of the design, such as computational methods for fragment library design, experimental and computational methods for fragment discovery and methods for the generation of structures of organic compounds. Examples are given of drug candidates, which were constructed using the fragment-based approach. The bibliography includes 156 references.
