Abstract: The data on the syntheses of serotonin 5-HT$_{6}$ receptor modulators published in recent years are surveyed. Modulators with high affinity to 5-HT$_{6}$ receptors with different selectivity degrees ranging from highly selective to semi selective and multimodal are described. The results of clinical trials of the most promising 5-HT$_{6}$ receptor modulators that arouse most interest in medicinal chemists are also described.
